Study on the inhibitory effect of Tangzhiqing extract on α-glucosidase in starch hydrolysis

Abstract
    [Objective] To investigate the inhibitory effect of Tangzhiqing(TZQ) on α-glucosidase involved in starch hydrolysis and the pathway of TZQ intervention in starch hydrolysis,focusing on each step of starch hydrolysis. [Methods] Using carbohydrates such as starch,dextrin,maltose,and isomaltose as substrates,the inhibitory effect of TZQ on α-glucosidase amylase,dextrinase,maltase,and isomaltase involved in starch hydrolysis was investigated using in vitro enzyme inhibition kinetics. [Results] Within the concentration range of 6.88×10-5 to 5.50×10-4 mg/mL,TZQ showed no dose-dependent inhibition of amylase activity;within the concentration range of 1.10×10-4 to 6.60×10-3 mg/mL,TZQ showed no significant inhibition of dextrinase activity;within the concentration range of 5.50×10-4 mg/mL to 1.10×10-2 mg/mL,TZQ showed a good inhibitory effect on maltase activity with an IC50 value of 2.96×10-3 mg/mL;within the concentration range of 5.50×10-4 mg/mL to 1.10×10-2 mg/mL,TZQ showed a good inhibitory effect on isomaltase activity with an IC50 value of 1.77×10-3 mg/mL. [Conclusion] TZQ can interfere with starch hydrolysis by inhibiting the activity of amylase,maltase,and isomaltase,thereby reducing blood glucose levels. TZQ is a potential α-glucosidase inhibitor.
