Influence of Danlou Prescription on the “dampness” in the early stage of coronary heart disease with blending of phlegm and blood stasis syndrome

Abstract
    [Objective] In this study,high fat diet combined with coronary artery left anterior descending artery ligation were used to establish a rat model of coronary heart disease(CHD) with blending of phlegm and blood stasis syndrome to observe the influence of Danlou Prescription(DLP) on the “dampness” in the early stage of CHD with blending of phlegm and blood stasis syndrome. [Methods] In this study,Wistar male rats were established a rat model of CHD with phlegm and blood stasis syndrome. At the end of 5 weeks,rats were randomly divided into 7 groups according to the ejection fraction(EF) after operation,including control group,sham surgery group,model group,DLP group low-dose group(200 mg/kg),DLP medium-dose group(400 mg/kg),DLP high-dose group(800 mg/kg),and atorvastatin group(10 mg/kg). At the end of 5 weeks,administration for 4 weeks(10 weeks),some samples were taken from rats in each group. Hemodynamics,cardiac morphology,blood lipid levels,and hemorheology,ischemic myocardial area(TTC staining),and myocardial pathological damage(HE staining) were measured to evaluate the establishment of a rat model of CHD with phlegm and blood stasis syndrome,and to observe the improvement of DLP on the “dampness” in the early stage of CHD with blending of phlegm and blood stasis syndrome. [Results] After model group fed with high-fat diet combined with coronary artery left anterior descending artery ligation,compared with control and sham surgery group,which the cardiac function was significantly decreased(P<0.01). Blood lipid levels were significantly increased(P<0.05 or P<0.01),and the blood was sticky,concentrated,condensed,and poly. And obvious infarct areas were observed after TTC staining of Model group rats myocardial tissue. After 4 weeks of administration,the cardiac function and structure of rats with CHD with phlegm and blood stasis syndrome were effectively improved,and the blood lipid levels decreased significantly,the myocardial injury degree of rats was lighter,the lesion range was smaller,interstitial edema and inflammatory cell infiltration were reduced. [Conclusion] DLP can effectively improve the heart structure and function of the “dampness” in the early stage of CHD with blending of phlegm and blood stasis syndrome. Reduce blood lipid level and improve the state of blood viscosity,concentration,coagulation and aggregation. Alleviate interstitial edema of myocardial tissue,and inhibit inflammatory cell infiltration.
