Abstract
|
[Objective] Comparison of the ameliorative effects of polydatin and verbenalin on immune function in cyclophosphamide(CY)-induced immunosuppressed mice.[Methods] Twenty-four male BALB/C mice were randomly divided into four groups:control group,model group,polydatin group and verbenalin group,with six mice in each group. The control group and model group were given pure water gavage,while the polydatin and verbenalin groups were given polydatin and verbenalin gavage treatment,respectively. After 11 days,the blood was collected,and serum immunoglobulin G(IgG) and immunoglobulin M(IgM) were measured by the enzyme-linked immunosorbent assay(ELISA). The thymus and spleen were isolated and weighed,then the thymus and spleen indices were calculated. The pathological structures of the liver,spleen,and thymus were analyzed by HE staining. RT-PCR was used to detect interleukin-2(IL-2) and interleukin-4(IL-4) expression in spleen. CD4+ and CD8+ T cell counts were assessed by flow cytometry. The proliferation of splenic B cells was evaluated using the CCK-8 assay.[Results] Consistently,after polydatin or verbenalin administration,the index of the spleen and the level of IL-2 both were significantly upregulated. The pathological alteration was also much improved. The lipopolysaccharide(LPS)-induced proliferation of B cells was much enhanced. In addition,after polydatin administration,the body weight and thymus index increased significantly. The levels of IgM and IgG and the counts of CD4+ and CD8+ T cells were also upregulated.[Conclusion] Polydatin and verbenalin have certain immunomodulatory effects,and the immunomodulatory effect of polydatin is better than that of verbenalin. |
