| Abstract

| [Objective] To establish the high-performance liquid chromatography chromatographic fingerprint and multi-component content determination method combined with chemometrics analysis,evaluate the quality of Nelumbinis Receptaculum from different regions. [Methods] An Agilent ZORBAX SB-C18 column(250 mm×4.6mm,5 μm) was used for the analysis,with mobile phase 0.2%(v/v) formic acid aqueous solution(A)-acetonitrile(B) gradient elution,and detection wavelength was 360 nm. The fingerprint of 16 batches of Nelumbinis Receptaculum from different regions was established and the content of six main components was determined simultaneously. The fingerprint similarity evaluation and chemometrics analysis were applied to evaluate the quality of Nelumbinis Receptaculum samples from different habitats. [Results] The similarity of fingerprints ranged from 0.984 to 0.999,and eight common peaks were calibrated. Among them,six components were identified,including hyperoside,isoquercetin,kaempferol-3-O-galactoside,isorhamnol-3-O-galactoside,isorhamnol-3-O-glucoside and quercetin. Both cluster analysis and principal component analysis classified 16 batches of Nelumbinis Receptaculum samples from different regions into two categories. Three marker components that affect the quality of the medicinal materials were screeded by orthogonal partial least square discriminate analysis,including isoquercetin,hyperoside and isorhamnol-3-O-galactoside. [Conclusion] The established fingerprint and multi-component content determination method is precise and stable which could be combined with chemometrics analysis to evaluate the quality of Nelumbinis Receptaculum. |
