| Abstract

| [Objective] To construct a new stable aqueous extract gel of Radix Codonopsis based on cationic Bletilla striata polysaccharide(CBSP) for anti-skin aging. [Methods] The aqueous extract of Codonopsis pilosula was prepared by water extraction. The effect of the aqueous extract of Codonopsis pilosula on the transcription activity of activated protein-1(AP-1) was studied to explore its anti-skin aging effect. A gel for local delivery of Codonopsis pilosula water extract was prepared. Firstly,cationic Bletilla striata polysaccharide was prepared by Williamson synthesis method,which was characterized by Fourier transform infrared spectrometer(FTIR),potential,degree of substitution of nitrogen(DS) and other tests. Then,the composite gel matrix(CBSP-CP) was prepared by electrostatic interaction with Carbopol(CP),and its steady-state viscosity,rheological curve and bio-compatibility were evaluated. CBSP-CP gel matrix and three kinds of CP gel matrices were combined with Codonopsis pilosula water extract respectively. [Results] the water extract of codonopsis pilosula can significantly inhibit the transcription activity of AP-1 and has certain anti-skin aging effect on skin. The results of FTIR,potential and DS show that CBSP has been successfully prepared. CBSP-CP composite gel matrix has steady-state viscosity and biocompatibility,and CBSP-CP Codonopsis pilosula aqueous extract gel has better stability in viscosity than three kinds of CP Codonopsis pilosula aqueous extract gels. [Conclusion] This study provides a reference for the development of highly stable gel of traditional Chinese medicine codonopsis pilosula extract,and also provides a way for the application of traditional Chinese medicine codonopsis pilosula extract in anti-skin aging products. |
