Bidirectional regulation of Dachengqi Decoction and Rhubarb on immunologic activity of macrophages |

Abstract
|
[Objective] To study the effect of Dachengqi Decoction (DCQ) and Rhubarb on the release of tumor necrosis factor α (TNFα) from normal or LPS-stimulated rat peritoneal macrophages (PMΦ). [Methods] TNFαwas assayed by MTT method, using the TNFα-sensitive cell line L929. [Result] The release of TNFαfrom PMΦstimulated by LPS could reach a peak after 8 hours, the peak was increasing with the LPS concentration increasing. Low concentrations of DCQ and Rhubarb could activate normal PMΦto release TNFαslightly. However, DCQ and Rhubarb evidently inhibited the excessive release of TNFαfrom PMΦstimulated by LPS, and the inhibition enhanced with the increasing of drug concentration. The inhibition mostly appeared in early phase after drug administration. [Conclusion] DCQ and Rhubarb have bidirectional regulation on the release of TNFαfrom macrophages. |
