| Abstract
|
| [Objective] To observe the effect of Scorpio extract on progranulocyte(HL-60) in acute promyelocytic leukemia and the correlative gene. [Methods] Scorpio induced morphologic changes of apoptosis of HL-60 and flow cytometry analysis, the effect on the distribution in cellular cycle of HL-60, the effect on the expression of modulating genes in apoptosis of HL-60 determined with immuohistochemistry were investigated. [Results] Scorpio could markedly accelerate the programming death of HL-60. The positive dose effect correlation could be found in certain concentration range. At the highest concentration(10 mg/mL) the proportion of apoptosis increased obviously, peaking at 21.44%. When middle or small dosage were administered, the proportion increased slightly. They were 12.56% and 9.47% respectively. It was only 7.36% in control. Simultaneously, it could markedly increase the expression of P53 gene(P<0.01 compared with control),and decrease the expression of bel 2. [Conclusion] Scorpio extract has beneficial effect on HL-60 death and correlative gene expression. |
