Protective Effect of HHI-I Against Phenylephrine-Induced Blood-Brain Barrier Permeability Breakdown in Rats |

Abstract
|
[Objective] To investigate the protective effect of HHI-I against the blood-brain barrier(BBB)permeability breakdown induced by phenylephrine in hypertensive rats.[Methods] Sixty male Wistar rats,weighing about 220 g were randomly divided into 6 groups with 10 animals in each.While the blank control group and model group were treated with nothing,three treating groups were received low,moderate and high dose of HHI-I(1.5%,3% and 6%,2 mL/kg respectively).The control group didn’t receive any treatment but equivalent normal saline.One hour after such intervention all these groups were intravenously injected with the indicator Evans blue,2%,4 mg/kg,b.w.Five minutes later the hypertension was induced in the last five groups with phenylephrine,0.01%,1.5 mg/kg,b.w.to result in BBB breakdown.Thirty minutes after that all the rats were killed and the cortex of hemisphere was harvested to measure the concentration of Evans blue in it.[Results] The BBB permeability was obviously increased in the rats of model group compared with the blank control(P<0.05).No change of BBB was observed in the N.S.control group.In contrast with those in model group,all the three doses of HHI-I could markedly decline the increased BBB permeability induced by phynylephrine.[Conclusion] HHI-I has significant protective effect against BBB permeability breakdown in hypertensive rats. |
