Abstract
|
[Objective] To observe the influence of astragalus injection on the proliferation and Na+-K+-ATPase activity of glomerular mesangial cells(GMC)cultured in high glucose or glycosylation end products(AGEs)medium.[Methods] GMC were incubated in culture medium containing high glucose or AGEs in the presence of astragalus injection and aminoguanidin for 48 hours.At the same time,the normal and control groups were added.The proliferation of GMC were analyzed by MTT method.The Na+-K+-ATPase activity were tested by Na+-K+-ATPase test Kit.[Results] GMC proliferated remarkably in glucose or AGEs medium in comparison with that in 5% FBS RPMI-1640 culture medium,and the Na+-K+-ATPase activity remarkably decreased.While the astragalus injection were added,the excess proliferation of GMC can be mostly reversed,and the Na+-K+-ATPase activity showed a significant restoration in comparison with that in control groups.[Conclusions] Astragalus injection can inhibit the excess proliferation and restore the decreased Na+-K+-ATPase activity of GMC induced by high glucose or AGEs,so it can protect the GMC. |
