Serum pharmacology of Qingdu suppositorium and its regulating effect on SiHa cell P53 gene expression in cervix cancer

Abstract
    [Objective] To investigate the regulating effect of Qingdu Suppositium on the growth of SiH a cell in cervical carcinoma and explore the mechanism with cytobiology and molecular biology.[Methods] Medicine-contained serum with different concentration was cultured with SiHa cell,and the blank serum and the positive control containing Chinese or Western medicine were established also.The influence on cell growth and the proliferation was determined with MTT.The cell cycle and apoptosis were examined with flow cytometry and the influence on the expression of cancer-inhibiting gene P53 was examined with PT-PCR semi-quantitative method.[Results] The cell quantity was reduced after exposure to the medicine-contained serum.Te serum with the concentration of 8% and the exposure time of 72 h had the most intensive effect.The number of cell in S phase was reduced obviously.The Qingdu Suppositium serum could accelerate SiHa cell apoptosis withou significant regularity; P53 gene expression of cells was significantly upregulated.[Conclusions] The serum containing Qingdu Suppositium can not only suppress directly the growth of cervical cancer cell but also inhibit tumor by regulating the P53 gene expression.
