Abstract
|
[Objective] To observe the effect of Zishui Qinggan decoction on hypothalamus ER-α mRNA expression and 5-HT content in rats with depression in peri-menopausal period.[Methods] Forty-five rats were randomly divided into five groups: control group,model group,TCM group,Estradiolum group and Estradiolum and Fluoxetine group.The rats of control group were treated with sham operation.The rats of other groups were treated with ovariosteresis and chronicity non-prediction stress.The rats of control and model group were administrated with physiologic saline and that of other group were individually administrated with Zishui Qinggan decoction,Estradiolum,Estradiolum and Fluoxetine.Hypothalamus ER-α mRNA expression and 5-HT content were determined by RT-PCR and HPLC-electrochemical detector method.[Results] The ER-α mRNA expression of model group was obviously lower than that of control group(P<0.01).The ER-α mRNA expression of TCM group was obviously higher than that of model group(P<0.05).The 5-HT of model group was obviously lower than that of control group(P<0.01) and The 5-HT of TCM group was obviously higher than that of control group(P>0.05).[Conclusions] The ER-α mRNA expression and 5-HT content exhibits low expression in rats of model group.Zishui Qinggan decoction may increase the content of 5-HT and increase the expression of ER-α mRNA and improve the condition of rats with depression in peri-menopausal period by enhancing ER-α mRNA transcriptional level and biological effect. |
