Abstract
|
[Objective] The acute myocardial infarction was established in rats.and then the rats were injected with endothelial progenitorcells (EPCs) preconditioned with Salvianolic acid B in different proportion combined with bone mesenchymal stem cells (BMSc) into theirheart ischemia area.The cardiac function of rats after cellular transplantation was observed.[Methods] The mononuclear cells were separated from rat bone marrow by density-gradient centrifugation using lymphocyte isolation and then the cells were seeded on fibronectincoated culture flask and purified.The acute myocardial infarction (AMI) was produced in rats by ligation of the left coronary arterythrough left thoracostomy.Different proportion of endothelial progenitor cells preconditioned with Salvianolic acid B combined with bonemesenchymal stem cells (BMSc) was injected into the five points of heart ischemia area.[Results] The structure of ventricles and cardiacfunction was showed enhanced in each cell transplanted groups.Compared with the control group, the left ventricular ejection fractionand the left ventricular cardiac output significantly increased in group 4:1 and 8:1 (P<0.05).[Conclusion] The (EPCs) preconditionedwith Salvianolic acid B combined with bone mesenchymal stem cells (BMSc) may increase the transplanted cell of myocardial infarctionarea in rat, improve the functions of left ventricle. |
