Protective effect of the silverweed cinquefoil root n-butanol fraction on the cardiomyocyte injured by hypoxia

Abstract
    [Objective] To study the protective effect of the silverweed cinquefoil root n-butanol fraction on the cardiomyocyte injured by hypoxia. [Methods] Myocardiocyte injured by hypoxia was established in rats in vitro. The metabolic activity and the survive rate of the myocardiocytes were determined respectively with MTT and Trypan blue stain. The activities of lactate dehydrogenase(LDH) and creatine kinase(CK) out of cells and superoxide dismutase(SOD) in cells,and the level of malondialdehyde(MDA) in cells were measured by spectrophotometry. [Results] Comparing with model group,the metabolic activity and the survival rate of cardiomyocytes were increased significantly in all groups treated by silverweed cinquefoil root n-butanol fraction(P<0.01),the releases of LDH,CK from myocardiocytes resulted from hypoxia were reduced(P<0.01),the activity of SOD was improved(P<0.01) and the production of MDA was inhibited in cells during hypoxia injury. [Conclusion] The silverweed cinquefoil root,n-butanol fraction exerts a remarkable protective effect on cardiomyocyte during hypoxia by improving the ability of clearing oxygen free radial in cardiac myocyte and inhibiting the oxide injury to membrane. It suggests that the silverweed cinquefoil root,n-butanol fraction can inhibit injury of cardiac muscles during hypoxia and this may be related with its anti-oxide ability.
