Effect of acupuncture on nucleus arcuatus INSR mRNA expression in rats with type 2 diabetes mellitus |

Abstract
|
[Objective] To study if the rats with Type 2 Diabetes Mellitus exist center insulin resistance and the effect of acupuncture on the expression of insulin receptor gene(INSR) of nucleus arcuatus in rats with Type 2 Diabetes Mellitus. [Methods] Obese rats due to diet were injected i.p. with small dose streptozotocin to induce type 2 diabetes mellitus. Then the rats were randomly divided into acupuncture, Glibenclamide and model group. After treatment for 4 weeks, FBS was examined with active blood sugar meter, FINS examined by radioimmunoassay, INSR mRNA expression of nucleus arcuatus were evaluated with hybridization in situ. [Results] Serum levels of FBS (15.79±1.87) and FINS (37.20±3.92) in model group were higher than that of control group (5.16±0.13, 23.57±1.63), amount of nucleus arcuatus positive cell (0.0416±0.0153) was significantly depressed compared with control group (0.1194±0.0242). Serum levels of FBS (5.46±0.20) and FINS(20.87±1.80) in acupuncture group were obviously lower than that of model group, amount of nucleus arcuatus positive cell (0.1524±0.0198) in acupuncture group was enhanced compared with model, which was similar to control. FBS (5.26±0.13) in Glibenclamide groups was reduced obviously compared with model, serum levels of FINS (28.10±1.86) and amount of nucleus arcuatus positive cell (0.0818±0.0234) in Glibenclamide group was improved when compared with model, but no significant differences were found between them. [Conclusion] Rats with Type 2 Diabetes Mellitus exists a depressed nucleus arcuatus INSR mRNA expression. Acupuncture and Glibenclamide can improve its expression of nucleus arcuatus in rats with Type 2 Diabetes Mellitus, but the effect of acupuncture was better than that of Glibenclamide. |
