| [Objective] To determine the content of Luteolin-7-O-β-D-glucoside and cosmosiin in Humulus scandens.[Methods] The content of humulus by Soxhlet was extracted and using high-performance liquid chromatography (HPLC) Humulus scandens in Luteolin7-O-β-D-glucoside and cosmosiin content were determined.The column was kromasilC18 and the mobile phase was methanol-0.2% phosphoric acid (45:55),the flow rate was set at 1.0 mL/min,the column temperature was 35 ℃,and the UV detector was set at 350 nm.[Results] Luteolin-7-O-β-D-glucoside content showed a good linear relationship within 0.05～0.40 μg (r=0.9997),the precision RSD= 2.06%,and the average recovery was 99.3%,RSD was 2.8%;cosmosiin content showed a good linear relationship within 0.072～0.36 μg (r=0.999 9),the precision RSD=1.73%,and the average recovery was 100.2%,RSD was 2.7%.The content of Luteolin-7-O-β-D-glucoside was the highest in July and August,and the cosmos was the highest in June.[Conclusion] The method is simple,sensitive,stable,reliable,and it can be used in the determination of Humulus scandens luteolin-7-O-β-D-glucosidase and the cosmos by HPLC.The appropriate harvesting season of Humulus scandens is from June to August. |
