Abstract
|
[Objective] To research the effects of Wu Yu decoction drug-containing serum on the expression of INHB,FSHR mRNA in ovarian granulose cell (abbr.GC) and the secretion function in rats. [Methods] Wu Yu decoction,FSH drug-containing serum and the normal rat serum were added into rat ovarian GC culture medium system. They were cultured for 48 hours. The expression of INHB was measured by ELISA,FSHR mRNA expression levels by RT-PCR,E2 levels of each group by radioimmunoassay. [Results] The E2 level was significantly higher in treatment groups than that in control groups,and the expression of INHB,FSHR mRNA significantly increased,among them medium dosage group of Wu Yu decoction was superior than that in Western medicine groups. [Conclusion] Wu Yu decoction drug-containing serum promotes the expression of INHB,FSHR mRNA and secretion of E2 of ovarian GC significantly. It is proved that Wu Yu decoction can enhance the ovarian reaction function to gonadotropins and improve the HPOA through adjusting internal factors of ovary and enhancing the expression of FSHR mRNA,thereby to promote the development and maturation of follicular and ovulation. |
