Effect of ginkgo biloba extract EGB761 on the mRNA expression of ACAT1 and LCAT in rabbit with Alzheimer’s disease

Abstract
    [Objective] To observe whether Ginkgo biloba extract EGB761 can influence the expression of ACAT1 and LCAT mRNA in rabbit with Alzheimer’s disease.[Methods] The male adult New Zealand rabbits were fed by 2% high cholesterol feed in order to establish the model of Alzheimer’s disease.EGB761 was used to treat the rabbits at a dose of 50 mg/(kg·d) for 3 weeks as EGB761 group.Simvastatin was fed at a dose of 5 mg/(kg·d) as a positive control drug.After intervention,the content of lecithin-cholesterol acyl transferase mRNA and cholesterol acyltransferase mRNA in hippocampus was tested by means of real-time quantitative RT-PCR.[Results] The content of ACAT1 mRNA in hippocampus was reduced by Ginkgo biloba extract EGB761,P<0.01 compared with model group.But the content of ACAT1 mRNA was not significant different compared with model group(P>0.05).[Conclusion] Ginkgo biloba extract EGB761 can down-regulate the expression of ACAT1 mRNA in rabbit with Alzheimer’s disease,however,it has no effect on the expression of LCAT mRNA.
