Abstract
|
[Objective] To observe the effect of the traditional Chinese medicine with benefiting qi and activating blood circulation on MMP-1 and TIMP-1 expression in rats with chronic heart failure(CHF).[Methods] We selected healthy male SD rats,established the model of heart failure by the way of coronary ligation accompanied with starvation and swimming.The rats were randomly divided into three groups: model group,Chinese medicine group,Western medicine group,and normal control group.After feeding it with drug or normal food for 6 weeks,the rats were killed and MMP-1,TIMP-1 gene expression and protein expression in myocardial tissue were detected using real-time quantitative fluorescent PCR technique and Western blot analysis(Western blot).[Results] Compared with the control group,the model group was higher than the normal group in MMP-1 expression(P<0.01),and TIMP-1 expression was significantly lower in normal group(P<0.01);the expression of MMP-1 of Chinese medicine and Western medicine group were lower than that in the model group(P<0.01),TIMP-1 expression was significantly higher than model group(P<0.01).[Conclusion] Yiqihuoxue Chinese medicine can lower the expression of MMP-1,increase the expression of TIMP-1,and thus prevent the damage of MMP-1 on the structure of the extracellular matrix function,delay myocardial remodeling and improve cardiac function,thereby prevent the development of CHF. |
