Study on differentiation of mesenchymal stem cells into cardiomyocyte-like cells induced by SalB in cardiac micro-environment

Abstract
    [Objective] To reproduce the model of myocardial ischemia-reperfusion injury to simulate cardiac micro-environment in vitro and observe the effect SalB in the process of mesenchymal stem cells(MSCs )differentiating into cardiomycyte-like cells. [Methods] A myocardium micro environment was simulated in vitro by adding myocardial cell lysate to MSCs culture system. MSCs was obtained from adult rats' bone marrow, and divided into four groups: the control group (group A), myocardio cell lysate group (TJ, group B), SalB group (group C) and TJ+SalB group (group D). They were cultivated for four weeks, and then the morphology was observed and the cTnT expression was detected by immune fluorescence detection technology. [Results] Each induced group had myocardial-like cell differentiation except group A. Compared with group C the cTnT expression in Group D was the most obvious with a statistical significance between them. [Conclusion] Myocardial cell lysate can induce MSCs differentiating into cardiomyocyte-like cells. Cardiac micro-environment can contribute SalB to induce differentiation of MSCs into cardiomyocyte-like cells.
