Effect of Shengji Xiangpi Gao on insulin-like growth factor-1 and its receptor in wound granulation tissue in rats with diabetic ulcer |

Abstract
|
[Objective] To observe the expression of insulin-like growth factor-1 (IGF-1) and its receptor (IGF-IR) content in the wound granulation tissue of rats with diabetic ulcer, explore the mechanism of Shengji Xiangpi Gao for promoting ulcer healing. [Methods] Using high fat diet plus tail vein injection with STZ preparation a model of type 2 diabetes was induced and perforating a hole in the back to make ulcer model. The rats according to the level of blood glucose and body weight randomly divided into blank group, vaseline group, Shengji Xiangpi Gao group. In 1, 7, 14, 21 d, 10 rats in each group were sacrificed, and fresh granulation tissue was obtained in 160 cases. Using real-time fluorescence quantitative PCR assay, the content of IGF-1 and receptor were determined [Results] The expression of IGF-1 increased at 7 d in Shengji Xiangpi Gao group, vaseline group, diabetic control group, reaching the peak at 14 days. In 21 d, the amount of IGF-1 in each group was obviously lower than that of blank group. In 7, 14 d, compared with vaseline group, diabetic control group it was P<0.05 in control group. Compared with blank group, Shengji Xiangpi Gao group had a increasing trend. In 21 d, compared with control group, IGF-1 decreased significantly in vaseline group, diabetic control group, Shengji Xiangpi Gao group (P<0.05). IGF-1R secretion in Shengji Xiangpi Gao group, vaseline group, diabetic control group in each time period was lower than the control group, and showed a decreasing tendency. In 7, 14, 21 d compared with control group, blank group and Shengji Xiangpi Gao group had significant difference. In 14, 21 d there was significant difference between Shengji Xiangpi Gao group and vaseline group. [Conclusion] IGF-1 increases in early and, middle period of diabetic ulcers, but decreases in the late. Shengji Xiangpi Gao can not promote the expression of IGF in diabetic ulcer granulation tissue, but can increase the expression of IGF receptor. |
