Determination of rutin content in Gynura divaricata by HPLC

Abstract
    [Objective] To establish assay method about the main ingredients of rutin in Gynura divaricata with HPLC. [Methods] Pretreatment was performed by petroleum ether (30~60℃). Mobile phase was “methanol-1% glacial acetic acid (40:60)”. The detection wavelength was 360 nm. Sample volume was 40 mL 1.000g 60% ethanol extract. Ultra extract was processed for 15 min. Chromatographic column was Xterra @RP 185 um. [Results] According to the linear relationship, it was showed that linear relation of rutin mass concentration was good in 0.011 2~0.067 2 mg/mL, r=0.999 1. All accuracy, reproducibility, stability and recovery could meet the analysis demand. [Conclusion] This method is simple and accurate, and it can provide some certain references for determination of rutin content in Gynura divaricata.
