Abstract
|
[Objective] To explore the influence of intervention at different time in MCAO rats by stimulating Neiguan acupoint with slow frequency. [Methods] The rats subjected to MCAO with suture method by Zea-Longa were randomly divided into seven group: normal group, sham operation group, model group, the non-acupuncture group, and stimulating Neiguan acupoint for a short time (5 seconds) group, stimulating Neiguan acupoint for a moderate time (60 seconds) group, stimulating Neiguan acupoint for a long time (180 seconds) group. The parameters of the three groups with simulation on Neiguan acupoint were fixed at 1 time/s, and the frequency was 5, 60, 180 seconds of acupuncture time respectively, and the acupuncture was performed once every 12 hours for 6 times. The effect indicators, including the normal animal nerve function, microcirculatory blood flow in the local area with ischemia, infarction rate of the brain tissue and the mechanism indicators, including the diameter of microcirculatory vascular input branches, diameter of the output branches, as well as the morphological indicators of the cells of brain tissue with HE staining were observed. In the model group observation was done immediately after modeling successfully, while in the later 4 groups the observation was performed 72 h after modeling. [Results] The improvement of the effect indicators in the three groups with simulation on Neiguan acupoint was better than the model group, the non-acupuncture group (P<0.05). Among the mechanism indicators the number of micrangium in cortex, hippocampus, striatum, the number of normal nerve cells (P<0.05) increased. As the improvement of each indicators the stimulation with a long time group was better. [Conclusion] Acupuncture on the Neiguan acupoint can get better effects. The acupuncture time can affect the Meridian effect. Under slow frequency the stimulation with longer acupuncture time can relatively get better effect; its intrinsic mechanism may be improving brain tissue microcirculation, regulating inflammatory reaction after ischemia and relieving the nerve cell necrosis. |
