Abstract
|
[Objective] To establish a method for simultaneous determination of asperosaponin VI and it's metabolites in rat bile by liquid chromatography-mass spectrometry (LC-MS/MS) and to study the excretion of asperosaponin VI from rat bile. [Methods] Bile samples were collected according to the designed time periods after intragastric administration. The content of asperosaponin VI and it's metabolites in rat bile were then analyzed using the established LC-MS/MS method. [Results] Asperosaponin VI and its three metabolites were simultaneously determined; asperosaponin VI and HN saponin were quantified at a higher concentration. Calibration curves were generated over a concentration range of 3.2~2 000 ng/mL for Asperosaponin VI and 0.8~500 ng/mL for HN saponin, respectively. Inter- and intra- day RSDs were both less than 7.72%. Biliary accumulated excretion for 24 hours after administration were 5 845.5 ng and 3 996.8 ng, respectively. This method is suitable for the determination of asperosaponin VI and its metabolite HN saponin F. [Conclusion] This method meets the requirements for the quantitative determination of biological samples and can be used for quantitative analysis of asperosaponin VI and its metabolite HN saponin F in rat bile. |
