Effect of Shengji Xiangpi plaster on expression of VCAM-1 and ICAM-1 in rats with diabetic ulcer in wound healing

Abstract
    [Objective] To investigate the mechanisms of the immune-inflammatory response promoted by Shengji Xiangpi plaster (SXP) in the process of ulcer healing in diabetic rats. [Methods] Wistar rats were randomly divided into four groups: one was served as normal control group and the remaining three groups rats were injected with STZ into tail vein for inducing diabetic. All the rats in this four groups were punched in the back for establishing the model of diabetic ulcer. The rats with diabetic ulcer in the three group were randomly stratified into diabetic control group, vaseline group and SXP group according to the blood glucose and body weight. The rats in SXP group were treated with SXP gauze compress externally, those in vaseline group treated with vaseline gauze compress and those in normal control group and diabetic control group were treated with physiological saline. The changes of VCAM-1 and ICAM-1 expression in the wound tissue were then observed. [Results] The diabetic ulcer wounds in the back of the rats in SXP group healed faster than those in the rats in diabetic control group and vaseline group. The difference was significant in 7th, 14th, 21th and 30th day compared with diabetic control group respectively. The expression of VCAM-1 in the rats in SXP group was lower than those in the rats in diabetic control group on the 14th, 21th and 30th day with significant difference. It was also lower than those in the rats in vaseline group on the 14th, 21th and 30th day with significant difference. The expression of VCAM-1 in rats of diabetic control group were higher than those in the rats of normal control group on the 21th and 30th day and the difference between them was significant; the expression of ICAM-1 in the rats in SXP group were lower than those in the rats of diabetic control group on the 14th, 21th and 30th day with significant difference; it were also significantly lower than those in the rats in vaseline group on the 21th and 30th day. The expression of ICAM-1 in the rats of diabetic control group were also significantly higher than those in the rats of normal control group on the 14th, 21th and 30th day. [Conclusion] Shengji Xiangpi plaster can promote the ulcer healing in diabetic rats, and it is closely related to regulating the immune-inflammatory response.
