Influence of Maxing Shigan decoction on expression of Sec14l3 in lung tissue of rats with asthma |

Abstract
|
[Objective] To study the influence of Maxing Shigan Gan decoction(MXSGD) and its main parts on the expression of Sec14l3 in the lung tissue of rat with asthma. [Methods] Seventy Wister rats were randomly divided into seven groups: normal group, model group, dexamethasone group, MXSGD group, Mahuang group, Xingren group, Gancao group, with 10 rats in each group. Rats were sensitized by intraperitoneal injection of ovalbumin on 0th and 7th day, followed by challenge with aerosolized ovalbumin on 14th day for two weeks and drugs administration. 24 h after allergen challenge, the inflammatory cells of BALF was tested. The inflammatory alteration of airway was observed through pathological slices and the expression quantity of the Sec14l3 mRNA in rat tissues was measured by RT-PCR. [Results] Compared with the normal group, the number of total cells, Eosinophils and Neutrophils in model group were increased obviously. The histopathological alteration of lung tissue exhibited serious inflammatory cell infiltration and airway epithelial injury. The Sec14l3 mRNA decreased substantially. Compared with model group, the number of total cells, Eosinophils and Neutrophils in MXSGD group, dexamethasone group, Gancao group decreased obviously. Seldom inflammatory cell infiltration and airway epithelial injury existed in lung tissue, and the Sec14l3 mRNA increased significantly, while Mahuang group and Xingren group had no difference with the model group. [Conclusion] MXSGD can effectively reduce the inflammatory cell infiltration and airway epithelial injury in the lung of rat with asthma, and the mechanism may be related to the regulation of Sec14l3 in the lung tissue and improvement of the protection against the airway epithelia. |
