Abstract
|
[Objective] Observating the effect of Schisandra Chinensis Mixture(SM) on the Wilms' tumor 1(WT1) protein and nephrin mRNA and podocin mRNA in renal of mice with ADN,to explore the mechanisms of SM decreases proteinuria. [Methods] Twenty-seven male Balb/c mice were randomly divided into three groups:normal control group, ADN group and SM group.The ADN was induced by a single tail-vein injection of ADR.The body weight, right kidney weight and UAER were measured. Furthermore, The morphological changes of renal tissue were observed under microscope. WT1 protein was observed by immunofluorescene.The mRNA of nephrin and podocin was determined by RT-PCR. [Results] Compared with the ADN group, there were significantly lower right kidney weight and UAER(P<0.05). There were significantly higher mRNA expression of nephrin and podocin in SM group than ADN group(P<0.01). WT1 was normal in control group,while it was focal absence in ADN group, and slightly absence in SM group. [Conclusion] After treatment with SM, WT1 protein, nephrin mRNA and podocin mRNA were up-regulated, which may safeguard podocyte, reduce proteinuria of mice with ADN. |
