Inhibitory effect of Yiqi Xiaozheng-contained serum on proliferation of EA.hy926 through intervention on ERmRNA and VEGFRmRNA expression |

Abstract
|
[Objective] To observe the effect of Yiqi Xiaozheng-contained serum on proliferation on EA.hy926 cells from umbilical vein of human and understand the way of inhibiting angiogenesis in human uterine myoma. [Methods] EA.hy926 cells cultured in vitro were divided into six groups: normal group, E2 group, E2 and ICI group,E2 and traditional Chinese medicine of high, middle, low dose group. EA. hy926 cells proliferation, migration ability, apoptosis rate, ERαmRNA, ERβmRNA and VEGFR1mRNA, VEGFR2mRNA expression were detected by MTT, scratch method and flow cytometry and RT-PCR technology respectively. [Results] Yiqi Xiaozheng-contained serum could blockade the promoting effect of E2 on the proliferation and migration of EA.hy926 decrease the expression of ERαmRNA,VEGFR1mRNA and VEGFR2mRNA, blockade the inhibitory effect of E2 on the apoptosis of EA.hy926 cells, up-regulate ERβmRNA expression with a significant difference when compared with E2 group (P<0.01). The effect and dose were dose-dependent. [Conclusion] Yiqi Xiaozheng-contained serum can inhibit proliferation and migration and promote apoptosis of EA.hy926 cells through down-regulating the expression of ERαmRNA, VEGFR1mRNA and VEGFR2mRNA and up-regulating the expression of ERβmRNA. Intervention of angiogenesis is one of the mechanisms for the treatment of uterine myoma. |
