Abstract
|
[Objective] To develop a UPLC method for simultaneous determination of five anthraquinones (Rhein,Chrysophanol,Emodin,Physcion and Aloe-emodin) in Radix et Rhizom a Rhei from different habitats. [Methods] The analysis was performed on Waters BEH C18 column (2.1 mm×150 mm,1.8 μm) with a gradient mobile phase of acetonitrile (A)-0.1% Formic acid solution (B) (0~3 min,19%→25%A,3~10 min,25%→100%A) at flow rate of 0.3 mL/min. The column was maintain at 30℃ and the detection wavelength was set at 254 nm. [Results] The linear ranges were 1.61~80.30 μg/mL for Rhein,4.98~249.20 μg/mL for Chrysophanol,3.27~163.30 μg/mL for Emodin,9.65~482.50 μg/mL for Physcion and 1.67~83.30 μg/mL for Aloe-emodin. The average recoveries of the five compounds were 99.7%,100.4%,100.1%,98.9%,100.9% and RSD were 1.5%,0.6%,0.8%,0.4%,1.7% respectively.[Conclusion] The rapid and accurate UPLC method was established. This method can be used to assess the quality of Rhubarb. |
