Neuroprotective role and its mechanism of Shenfu injection on focal ischemic stroke in rats |

Abstract
|
[Objective] To investigate the effect of Shenfu injection on the expression of Na-K-Cl-cotransporter 1 (NKCC1) in rats with focal cerebral ischemic stroke. [Methods] The ischemic stroke was generated by 90 min occlusion of right middle cerebral artery. After 24 h reperfusion, neurological score was evaluated. The cerebral infarct volumes were quantified by 2, 3, 4-triphenytetrazolium-chloride (TTC), and the NKCC1expression were evaluated by Western blot and immunofluorescence methods. [Results] Shenfu injection-treatment group (SFI) significantly reduced the infarct volume (P<0.05) and the degree of neurological deficits (P<0.05). Compared to the ischemia group, the protein expression of NKCC1 was significantly reduced in the SFI group. [Conclusion] Shenfu injection plays a neuroprotective role in rats with experimental cerebral ischemia, which was associated with the inhibition of NKCC1. |
