Abstract
|
[Objective] To observe the preventive effect of Xiaozhi capsule against atherosclerosis in rabbits, and provide some experimental evidences for further study. [Methods] Animal were divided into control group, model group high dose group of Xiaozhi capsule and and low dose group of Xiaozhi capsule. At the end of treatment (10 weeks after dosing), ear vein blood was obtained for testing superoxide dismutase (SOD), malondialdehyde (MDA), nitric oxide (NO), adiponectin (ADP), C-reactive protein (CRP) and endothelin-1 (ET-1) content; simultaneously blood pressure was measured;the sample of heart, aorta, lower extremity arteries and liver were also harvested, fixed by 12% formaldehyde solution, and the histopathology was examined, scoring on the animal arteriosclerosis and analyzing the arterial stiffness index. [Results] Xiaozhi capsule was administered for 10 weeks. The high dose group of Xiaozhi capsule could significantly increase the activity of SOD in serum of rabbits. High and low dose groups of Xiaozhi capsule could significantly increase the rabbit serum level of ADP, decrease MDA, CRP and ET-1, decrease systolic blood pressure, and low dose group of Xiaozhi capsule could decrease the diastolic blood pressure. The histopathology showed that high and low dose groups of Xiaozhi capsule could alleviate lesion of atherosclerosis in myocardial arterioles and vacuolar degeneration of hepatocytes. [Conclusion] Xiaozhi capsule could regulate effectively the index of atherosclerotic lesions in rabbit, adjust the blood pressure and scavenge free radicals, prevent lipid peroxidation, reduce inflammation medium and protect vascular endothelial cell. It was showed that the drug has good control effect in the occurrence and development of rabbit artery atherosclerotic lesions. The research results of the drug in the clinical prevention and treatment of atherosclerosis have provided some experimental basis. |
