Preliminary research on the role of TLR4 at acupoints in the inflammatory reaction produced by acupuncture

Abstract
    [Objective] To investigate the role of TLR4 at acupoints in the inflammatory reaction produced by acupuncture.[Methods] The 80 health rats were divided into acupuncture group and blank control group, each group 40 rats. Each group was randomly divided into 8 time points(immediate, 15 min, 30 min, 1 h, 2 h, 4 h, 8 h, 24 h), 5 rats per point. Testing the concentrations of TLR4, HMGB1, IL-1α, IL-6, TNF-βin point area including skin and muscle tissue with ELISA. [Results] There were no significant changes in the concentration of each index at each time point in control group. Acupuncture could increase the concentrations of TLR4, HMGB1, IL-6, TNF-β, whereas IL-1αconcentrations did not change. The concentrations of HMGB1, TNF-βhad certain relativity with the concentration of TLR4 respectively after acupuncture. However, the concentration of IL-6 had weaken relativity with TLR4and the concentration of IL-1αhad uncertain relativity with TLR4. [Conclusion] TLR4 located at acupoints may play an important part in the inflammatory reaction produced by acupuncture.
