Effect of effective components of Chinese medicine for tonifying kidney on the levels of oxidative stress in SAMP8 mice

Abstract
    [Objective] To observe the effect of effective components in tonifying kidney herbs of psoralen, oleanolic acid, icariin on oxidative stress level with mice (SAMP8). [Methods] This experiment chose september-aged male SMAP8 mice, according to the random number table method were randomly divided into psoralen group, oleanolic acid group, icariin group and model group. At the same time another age segment SAMR1 mice were chosen as control group. The treatment group were given psoralen (50 mg/kg), oleanolic acid (50 mg/kg), icariin (20 mg/kg) by gavage, saline control group and model group were given the same amount of gastric perfusion, 0.3 mL each time, 1 time a day, continuous intragastric administration for 4 weeks, then the rats were killed by decapitation and getting the brain. Using enzyme-linked immunosorbent assay (ELISA) method for detecting the contents of glutathion peroxidase (GSH-Px), catalase (CAT), superoxide dismutase (SOD) in cerebral cortex of SMAP8. [Results] SOD, CAT and GSH-Px activity in SAMR1 mice were decreased compared with age-matched of SMAP8 in the cerebral cortex; cerebral cortex SOD, CAT and GSH-Px activity in the model group were lower than the control group, after 4 weeks of treatment, SOD, CAT and GSH-Px activity in the cerebral cortex of the treatment group were higher than those in the model group, and there was no significant difference between psoralen, oleanolic acid, icariin three groups of the above indicators (P>>0.05). [Conclusion] The kidney tonifying herbs has certain protective effect on oxidative stress injury in SAMP8, so as to achieve the effect of prevention and treatment of senile dementia.
