Effect of Laifu Chengqi decoction on the apoptosis of renal cells in rats with severe acute pancreatitis |

Abstract
|
[Objective] To investigate the effect of Laifu Chengqi decoction on the apoptosis of renal cells and expression of apoptosis gene Bax and Bcl-2 in rats with severe acute pancreatitis. [Methods] The 72 healthy Wistar rats were randomly divided into control group (24 rats), SAP group (24 rats) and Laifu Chengqi decoction group (24 rats). Heart blood of rats in the three groups was taken at 6 h, 12 h, 24 h after operation to detect serum Cr and BUN. Renal cells stained by Annexin V- FITC/PI were drawn off from kidney to detect apoptosis rate using flow cytometer. The expression of Bax and Bcl-2 mRNA in pancreatic tissue were detected by RT-PCR. [Results] Compared with the control group, with the change of time, serum Cr and BUN in SAP group were increased significantly (P<0.01). Index of renal cell apoptosis was increased gradually. The expression of Bax mRNA was significantly increased(P<0.01). The expression of Bcl-2 mRNA was significantly decreased(P<0.01). Compared with the SAP group, serum Cr and BUN in Laifu Chengqi decoction group were decreased (P<0.01), and index of renal cell apoptosis was decreased. The expression of Bax mRNA was significantly decreased (P<0.01), the expression of Bcl-2 mRNA was significantly increased (P<0.01). [Conclusion] Laifu Chengqi decoction may regulate the apoptosis related gene to decrease the apoptosis of renal cells, thereby reducing renal pathological damage. |
