| Abstract
|
| [Objective] To observe the effect of electro-acupuncture on behavior change and the cytokines of IL-1β, IL-6, TNF-α in the spleen of sleep deprivation rats to investigate the mechanism of electro-acupuncture treatment in insomnia. [Methods] Thirty-two Wistar rats with SPF level were randomized into the control group, model group, diazepam group and electro-acupuncture group. The sleep deprivation rats were established by DL-P-Chlorophenylalanine (PCPA) abdominal injection. The diazepam group were treated by diazepam, and the electro-acupuncture group were treated by electro-acupuncture with Baihui (GV20) and Shenting (GV24) acupoints. Behavior change and the cytokines of sleep deprivation rats were observed and analyzed by real time-PCR after 5 days continuously treatment. [Results] Comparison with the control group in the open field test, the crossing scores, the rearing scores and the pace time in center of the rats were significantly up-regulated in the model group(P<0.05). On the other hand, all of those scores mentioned above were significantly increased in the group of diazepam and electro-acupuncture(P<0.05). In the results of real time-PCR, the IL-1β and the TNF-α expression levels were remarkably higher in the model group than in the control group(P<0.01). Furthermore, the IL-1β and the TNF-α expression levels in the diazepam group and the electro-acupuncture group were Significantly down-regulated(P<0.05). But there was no obviously results in each group with the level of IL-6 (P>0.05). [Conclusion] The electro-acupuncture can effectively treat the symptoms of Insomnia, and it also can proper control the level of cytokines. |
