Effects of Shenjin Yigu orthopaedic manipulation on chondrocytes metabolismand the expression of Sox9 in rabbits

Abstract
    [Objective] To study the effects of Shenjin Yigu orthopaedic manipulation on rabbit chondrocytes metabolism and the expression of Sox9.[Methods] KOA rabbits were randomly divided into 2 groups.The treatment group used Shenjin Yigu orthopaedic manipulation therapy, the control group using intra-articular injection of sodium hyaluronate, culturing chondrocytes in vitro after drawing materials.Microscope observation and comparing of chondrocytes' proliferation between two groups by MMT method, analyzing the expression of Sox9 of chondrocytes between two group by Western-blot method.[Results] Microscopic observation of the treatment group and control group of chondrocytes proliferation faster than those in the blank group and the multiplicaion rate of the treatment group was fastest than those in the control and blank groups, and better proliferation of chondrocytes in the treatment group than that in the control group detected by MMT method(P<0.05).Western blot analysis showed that the expression of Sox9 was higher in the treatment group than that in the control group(P<0.05).[Conclusion] Shenjin Yigu orthopaedic manipulation can improve the speed of proliferation of chondrocytes, enhancing chondrocytes' multiplication ability.Simultaneously this manipulation can increase the expression level of Sox9 in injured cartilage tissue, to promote the synthesis of chondrocyte metabolism ability.
