Effects of Shenzhujing tablets for sodium pentobarbital-induced hypnosis in mice |

Abstract
|
[Objective] To observe the effects of Shenzhujing tablets on sodium pentobarbital-induced hypnosis. [Methods] The 96 of Kunming mice were randomly divided into 4 groups: normal group, and Shenzhujing tablet (0.5, 0.25, 0.125 g/kg) groups. The drugs were administrated orally in single doses to 12 of the mice in each group 2 h before the intraperitoneal injection of sodium pentobarbital at the dose of 30 mg/kg or 50 mg/kg, respectively. Then the mice received daily oral treatments of drugs for 7 days. Similarly, Sodium pentobarbital-induced hypnosis tests were also used 2 h after the last administration of the drugs. The sleep latency and sleep duration during pentobarbital-induced sleep were measured and the ratios of sleep mice to non-sleep mice were counted in mice after oral administration of the drugs. Moreover, the contents of the 5-hydroxytryptamine(5-HT) and 5-hydroxyindoleacetic acid(5-HIAA)in the brain stem were determined. [Results] Compared with normal group, Shenzhujing tablets significantly prolonged the pentobarbital-induced sleep duration and decrease sleep latency at dose of 0.5 g/kg(P<0.05), increase the ratios of sleep mice(P<0.01) to non-sleep mice and the contents of 5-HT in the brain stem under the condition of 30 mg/kg of sodium pentobarbital (P<0.05). [Conclusion] Shenzhujing tablets possess sedative-hypnotic activity possibly by promoting the secretion of 5-HT in the brain and might be effective agents for treatment of insomnia. |
