Experimental study of Gegen Qinlian Tang on the impact of early retinopathy in diabetic rats |

Abstract
|
[Objective] To investigate the therapeutic effect of Gegen Qinlian Tang early stage of diabetic retinopathy in rats.[Methods] The 45 Wistar rats, streptozotocin (STZ) intraperitoneal injection, making diabetes were randomly divided into DM group, DM+NS group (saline injection group diabetes diabetic model) and the experimental group (DM+Gegen Qinlian group). After modeling, DM group received no treatment, and the experimental group was given Gegen Qinlian 8.2 g/kg, and DM+NS group was injected with an equal volume of saline. Diabetic rats, fasting blood glucose, plasma insulin, body weight, triglyceride (TG) and high density lipoprotein (HDL-C) content were detected to observe Gegen Qinlian hypoglycemic lipid-lowering efficacy; by Western blot method retinal protein kinase B (protein kinaseB, PKB) and inflammation-related factors between a cell adhesion molecule 1 (intercellular adhesion molecule1, ICAM-1) expression; HE staining used ultrastructure of rats in each group; with TUNEL assay of apoptosis-related protein C-myc in the apoptotic index.[Results] Compared with DM group and DM+NS, the experimental group was significantly lower fasting blood glucose, body weight increased significantly, statistically significant difference (P<0.05); the DM and DM+NS group, the experimental group of rats blood insulin content increased, the difference between groups was statistically significant (P<0.05); the DM and DM+NS group, the rats in the experimental group was significantly lower TG and HDL-C was significantly increased (P<0.05). By Western blot detection of visible and DM and DM+NS group, and PKB expression in rat experimental group increased significantly, and ICAM-1 expression was significantly decreased. The difference was statistically significant (P<0.05). HE staining was observed under light microscope ultrastructure groups:DM group pyknosis, stain, nuclear membrane retraction, chromatin margination, apoptotic bodies, changes in the experimental group was significantly reduced (P<0.05). The DM and DM+NS group, the experimental group of apoptosis-related protein C-myc decreased (P<0.05).[Conclusion] Gegen Qinlian Tang can improve fasting blood glucose, plasma insulin, and the change of body weight, with good blood glucose and lipids, by increasing the expression of PKB and suppress inflammation in diabetic rat retinopathy to reduce or delay the occurrence and development of diabetes. |
