Study on the podocyte associated protein α-actinin-4 of experimental diabetic rats treated with Zhenhua Xiaoshen An capsules

Abstract
    [Objective] Through this experiment to observe the experimental diabetic rats renal and podocyte associated protein α-actinin-4 expression, indicate Zhenhua Xiaoshen An capsules effectiveness of diabetic nephropathy in the treatment of diabetic nephropathy.[Methods] To establish the model of STZ induced diabetic rats model group, control group, Traditional Chinese medicine group, Western medicine group. Traditional Chinese medicine group was given Zhenhua Xiaoshen An capsule. Western medicine group was given irbesartan. Fasting blood glucose, 24 h Urine micro albumin, Scr, BUN were detected with optical microscope, transmission electron microscope and immunohistochemistry in order to observe the expression of podocyte skeletal protein α-actinin-4. The expression of α-actinin-4 mRNA were detected with Real-time quantitative PCR.[Results] After the intervention, the urine volume of traditional Chinese Medicine traditional Chinese medicine high, middle, low dose group and Western medicine control group obviously decreased, compared with that of DM model group (P<0.01). The blood glucose of rats in Traditional Chinese medicine every dose group and Western medicine control group were obvious lower than that in DM model group (P<0.01). The α-actinin-4 mRNA contents of rats in traditional Chinese medicine every dose group and Western medicine control group were lower than that in DM model group (P<0.01).[Conclusion] Zhenhua Xiaoshen An capsules can reduce to experimental DM rats blood glucose, serum creatinine, blood urea nitrogen and excrete urine micro albumin, and improve renal function. It also can restrain the incresing of the renal volume of experimental DM rats at early stage and the protein and mRNA expression of α-actinin-4.
