Abstract
|
[Objective] A UPLC-Q-TOF-MS method was developed for the analysis of absorbed components in SD rat plasma after oral administration of Lianhua Qingwen capsules (LQC) for the first time.[Methods] The blood samples were taken from the posterior orbital venous plexus of rats and methanol was used for the protein precipitation.The separation of components was achieved on a C18 column by gradient elution with methanol -0.1% formic acid water solution as mobile phase.Comparative analysis of blank plasma,mixed reference standards and plasma with LQC was performed by UPLC-Q-TOF-MS in negative ion mode.[Results] The results suggested that 22 prototype compounds were identified,including flavonoid,anthraquinone,phenylpropanoid and iridoid.These compounds were identified and confirmed by comparing their retention times,mass spectrometry fragmentation characteristics with that of references as well as literatures.[Conclusion] The study of the absorbed components of LQC in plasma provides an experimental basis for revealing the effective components and its pharmacokinetics study. |
