Study on the effects of endometrium of women with polycystic ovary syndrome by Bushen Tiaochong decoction

Abstract
    [Objective] Study on the effects of Bushen Tiaochong decoction on endometrium of women with polycystic ovary syndrome. [Methods] In vitro cultivation human endometrium cells of women with polycystic ovary syndrome. Endometrial cells were be divided into the experimental group treated with Bushen Tiaochong decoction, positive control group treated with progesterone, negative control group treated with 17 beta estradiol and the blank control group. Observation the ultrastructure changes of endometrium cells after drugs intervention by transmission electron microscope. Assay the expression of progesterone receptor in endometrial cells for each group by ELISA. Detect the expression of TGF-β1 and Smad4's mRNA by semi-quantitative RT - PCR and the expression of TGF-β1 and Smad4's protein by Western blot. [Results] Compared with the estrogen group and the control group, the quantity of microvilli on cells' surface and the volume of mitochondria of the progesterone group and the experimental group increased. The rough endoplasmic reticulum and the Golgi apparatus of the progesterone group and the experimental group swelled. The concentration of progesterone receptor in endometrial cells of experimental group was lower than that of the estrogen group and the control group (P<0.05). [Conclusion] Bushen Tiaochong decoction can promote the transformation of endometrium from proliferative phase to secretive phase.
