Exploring the antipyretic mechanism of Airening suppository by rectal drug administration

Abstract
    [Objective] To observe the antipyretic mechanism of Airening suppository. [Methods] Rabbits were randomly divided into blank control group, model control group, Airening suppository group Xiaoyantong suppository group. The models of fever induced by 2,4-binitro-phenol, and measured temperature at different times after the treatment. The IL-6, COX-2, PGE2, AVP, α-MSH were detected in serum of each rabbit by ELISA, and Immunohistochemistry is used to detect the expression of NF-κB of the liver. [Results] After 0.5 h, compared with model control group, the body temperature of experimental groups started to descend (P<0.05), and body temperature descended significantly after 1 h (P<0.01). The contents of COX-2 and PGE2 were decreased significantly (P<0.05), and the contents of AVP and α-MSH were increased significantly (P<0.05). The contents of COX-2 and PGE2 were decreased significantly (P<0.05). The expression of liver NF-κB decreased significantly (P<0.05). The contents of IL-6, COX-2 and PGE2 were decreased significantly of Airening suppository group(P<0.05), and the contents of AVP was increased obviously (P<0.05). The expression of liver NF-κB significantly lowered (P<0.05). [Conclusion] Airening suppository can significantly reduce the body temperature of rabbits, and the curative effect is equivalent to Xiaoyantong suppository. The antipyretic mechanism of Airening suppository may be related to the regulation of NF-κB pathway. It suggests that Airening suppository has a antipyretic function with multiple targets.
