Early intervention of storax promotes long-term neurological function restoration after ischemic stroke in rats

Abstract
    [Objective] To investigate whether early intervention of storax promotes long-term neurological function restoration after ischemic stroke in rats.[Methods] Healthy male SD rats were randomly divided into vehicle and storax groups. Rats were subjected to 2 hours of occlusion of middle cerebral artery (MCAO), then administered with storax intragastrically at 1, 3, 6 hours after MCAO for 3 times. Modified neurological severity scores (mNSS), adhesive tap-removal test and foot fault test were performed at 1, 4, 7, 14, 28 days after MCAO to evaluate neurological function, at the same time, weight and death rate were recorded. Hematoxylin eosin (HE) staining was performed to observe repairmen of brain tissue, and immunofluorescence staining of platelet endothelial cell adhesion molecule-1 (CD31) and synaptophysin (SYP) was also performed.[Results] The weight of rats were decreased at 1, 4 days after MCAO and gradually increased from 7 days to 28 days after MCAO, compared vehicle group. Weight reduction was smaller and weight incretion was bigger in storax group, but there was no significant difference between the two groups. The mNSS scores, the tap-removal time and the foot-fault steps were increased at 1 day and gradually decreased till 28 days after MCAO, storax decreased the mNSS scores, the tap-removal time and the foot-fault steps at the above time points, and significantly decreased the mNSS scores at 21, 28 days and foot-fault steps at 4, 7, 14 days after MCAO (P<0.05). HE staining showed that brain tissue in storax group got a better repairment than that of vehicle group. Immunofluorescence staining showed that CD31 and SYP positive expression in ischemic border were significantly increased in storax group (P<0.05) when compared with vehicle group.[Conclusion] Early intervention of storax promotes long-term neurological function restoration and neurovascular remolding after stroke in rats.
