Effects on protein expression of Fas, Fasl and Caspase-8 in alcohol liver disease of rats intervented by Wutian Baogan Ye

Abstract
    [Objective] To investigate the Wutian Baogan Ye's effect on the apoptosis of liver cells, and the expression of Fas, Fasl, Caspase-8 in liver tissue of alcohol liver disease (ALD) rats.[Methods] The 100 Wistar rats were divided into six groups randomly:normal group (10), model control group(18),Wutian Baogan Ye low dose group(18), middle dose group(18), high dose group(18), positive drug group(18). Rat model of ALD was induced by perfusing stomach with liquor. After 12 weeks, rats were anesthesia by diethyl ether and decapitation to execute. Hepatic tissue of rats was superscription. Index of apoptosis was detected by TUNEL. Expression of Fas, Fasl in liver tissue of rats were observed by immunohistochemisty.[Results] Compard with normal rats, the index of apoptosis was higher in model group, and Wutian Baogan Ye group (middle and high dose) and positive drug group was lower than that in the model group. There was no significant difference between low dose group and model group in the index of apoptosis. Expression of Fas was not different among groups. The expression of Fasl and Caspase-8 in model group was significant higher than other groups. Compared with model rats, Wutian Bangan Ye (middle and high dose) groups and positive drug can decreased the expression of Fasl and Caspase-8. Wutian Baogan Ye low dose group decreased the erpression of Fasl but had no statistical significance compared with model group.[Conclusion] Wutian Baogan Ye had an active effect on inhibiting apoptosis of liver cell in ALD through decreased the expression of Fasl, Caspase-8 to treat and protect from ALD.
