Effect of Luqi formula on blood pressure, angiotensin-aldosterone and myocardial collagen fibers in rats with hypertension complicated with heart failure

Abstract
    [Objective] To study the effect of Luqi formula on blood pressure, angiotensin I, angiotensin Ⅱ, aldosterone and myocardial collagen fibers in rats with hypertension complicated with heart failure.[Methods] The rat models of hypertension complicated with heart failure was induced by abdominal aorta constriction. After successful modeling, the rats were randomly divided into model group, the control group, the sham operation group, the high, middle and low dose of Luqi formula groups. After 8 weeks of administration, measurement of blood pressure in rats. Determination of angiotensin I (AngI), angiotensin Ⅱ(AngⅡ) and aldosterone (ALD) levels in rats with ELISA double antibody sandwich enzyme. Picrosirius red staining was used to observe the changes of myocardial collagen fibers in rats.[Results] Compared with the model group, each dose of Luqi formula can effectively reduce systolic blood pressure in rats with heart failure(P<0.05). The high and middle dose gruops were better than the low dose group(P<0.05). Compared with the model group,each dose group of Luqi formula can effectively reduce AngI, AngⅡ and ALD(P<0.05). The high dose group of Luqi formula was obviously better than the low dose group in reducing ALD(P<0.01). Through the picrosirius red staining of rats'myocardium,each dose group of Luqi formula compared with model group can significantly reduce the myocardial collagen content,improve myocardial interstitial morphology.The high dose group of Luqi formula was better than the middle and low groups.[Conolusion] Luqi formula can reduce blood pressure of heart failure rats, regulate neuroendocrine activation, reduce myocardial fibrosis, improve ventricular remodeling and reduce myocardial damage.
