| Abstract

| [Objective] To improved extraction process of lumbrokinas.[Methods] Lumbrokinas was preparation from fresh earthworm by extraction, separation and purification. Sequential comparison of four solvent extraction methods, four precipitation separation methods and two chromatography purification methods, to obtaine a better process by investigating enzyme activity. Then the process was improved by fractional precipitation, removal of impurities and gel chromatography.[Results] Lumbrokinas was preparation by optimization of the extraction process, including buffer extraction, ethanol fractionation precipitation, DEAE-Sepharose FF and Sephadex G-75 chromatography. The enzyme activity was 28 000 μ/mg, which was higher than that of the old process.[Conclusion] The improved process has high enzyme activity and simple operation, and is suitable for industrial production. |
