Abstract
|
[Objective] To study the effects of the combination of emodin and tanshinol in hepatic fibrosis rats with carbon tetrachloride (CCl4).[Methods] Fifty male SD rats were randomly divided into normal group, model group, emodin group, high dose emodin and tanshinol combination group, low dose emodin and tanshinol combination group. The hepatic fibrosis rat models in each group were induced by intraperitoneal injection of CCl4 except normal group. Starting from the third week, in addition to injection molding, each experimental group was given corresponding drug gavage respectively, the normal group and model group were given intragastric administration of physiological saline for 6 weeks. After the last administration, blood and liver tissue were taken to observe the histopathological changes of the liver. The activities of serum alanin aspartate aminotransferase (AST), aminotransferase(ALT) and alkaline phosphatase(ALP) were measured. Levels of laminin (LN), hyaluronic acid (HA) and procollagen Ⅲ(PCⅢ) in serum were detected by enzyme linked immunosorbent assay (ELISA). The content of hydroxyproline (HYP) in liver homogenate was measured by acid hydrolysis method.[Results] Compared with the model group, the levels of ALT, AST, ALP, LN, HA, PCⅢ and HYP liver tissue of each experimental group was significantly decreased(P<0.05, P<0.01), the liver pathological fibrosis was obviously relieve. Compared with emodin group, the levels of ALT, AST, ALP, LN, HA and HYP in high dose emodin and tanshinol combination group were significantly decreased (P<0.05 or P<0.01).[Conclusion] Emodin has antifibrotic effect; tanshinol can enhance the effect of emodin on antifibrotic effect. |
