Protective effect and mechanism of volatile oil from Yuxingcao on cerebral ischemia-reperfusion injury in mice

Abstract
    [Objective] To explore the protective effect and mechanism of volatile oil from Yuxingcao on cerebral ischemia reperfusion injury in mice. [Methods] one hundred and twenty SPF C57BL/6 mice were randomly divided into six groups, including Sham-operated group (20), model group (20), simvastatin group (XFTT, 20), low-dose volatile oil of Yuxingcao group (YXC-L, 20), middle-dose volatile oil of Yuxingcao group (YXC-M, 20) and high-dose volatile oil of Yuxingcao group (YXC-H, 20). Fourteen days lavage was done after prophylactic administration. The dose of XFTT group, YXC-L group, YXC-M group and YXC-H were 20 mg/(kg·d), 5 mg/(kg·d), 10 mg/(kg·d) and 20 mg/(kg·d) separately, which given by lavage administration. Sham-operated group and model group gave the same dose of sodium chloride. 1h after last administration, MCAO model was builded in those groups except for Sham-operated group. After the experiment, the change of behavioral evaluation, cerebral infarction area, cerebral water content and hippocampal dentate gyrus were observed, the activity of superoxide dismutase (SOD) and the content of tumor necrosis factor-α(TNF-α) and interleukin-8(IL-8), malondialdehyde (MDA) in serum were measured. [Results] Compared with model group, behavior score and morphological change of neurons in hippocampal dentate gyrus were obviously improved in YXC-L group, YXC-M group and YXC-H group. and decreased cerebral infarction rate, cerebral water content, the content of TNF-α, IL-8 and MDA(P<0.05), while the activity of SOD were increased(P<0.05). which appeared dose-dependent. The above index were improved in XFTT group. [Conclusion] Volatile oil from Yuxingcao has protecting effects on MCAO model mice, and appear dose-dependent.
