Andrographolide inhibits lipid droplet accumulation by blocking PPARγ-C/EBPα expression

Abstract
    [Objective] To investigate the inhibitory effect of andrographolide on adipocyte differentiationand lipid droplet accumulation.[Methods] Different concentrations of andrographolide were added to 3T3-L1 adipocytes. The effect of andrographolide on the formation of fat droplets was performed by highcontent imaging. The molecular mechanism of andrographolide was illustrated by detecting the expression of PPARγ and C/EBPα.[Results] Andrographolide reduced lipid accumulation in dose-dependent manner. The expression of PPARγ and C/EBPα were significantly reduced at 30 μmol/L (P<0.01) without obvious cytotoxicity (P >0.05).[Conclusion] Andrographolide inhibited adipocyte differentiation through blocking PPARγ-C/EBPα expression and reduced excessive lipid droplets accumulation, which holds potential for alleviating obesity.
