Effects of Fushen recipe on expression of TGF-β1, E-cadherin and α-SMA in peritoneal mesenchymal membranes of uremia rats |

Abstract
|
[Objective] To invigorate the kidney in uremic rats TGF-β1, expression of E-cadherin and α-SMA.[Methods] SD rats underwent 5/6 +1.5% renal dialysis Baxter replication in rat model of peritoneal fibrosis. After 6 weeks' intervention, the serum levels of creatinine and urea nitrogen were detected. The pathological changes of peritoneum were observed by HE staining. The expression of E-cadherin and α-SMA in peritoneal mesothelial markers in each group was detected by RT-PCR method, and the content of TGF-β1 was detected by ELISA.[Results] The results help kidney prescription intervention, serum creatinine and urea nitrogen were decreased, especially in the high dose group was most significant (P<0.05); Fushen recipe Group of peritoneal mesothelial cell degeneration, shedding, inflammatory cell infiltration and neovascularization were obviously reduced; the results of RT-PCR showed that E-cadherin decreased in model group compared with normal group the expression, the difference was statistically significant (P<0.05); TGF-β1 and α-SMA compared with normal group expression, the difference was statistically significant (P<0.01), compared with the model group, the intervention group to help kidney. The expression of E-cadherin was increased, and the high dose group increased most significantly (P<0.01); TGF-β1 and α-SMA in middle and high dose groups had statistically significant difference (P<0.05, P<0.01); the other group compared with control group, low dose group decreased, the results were not statistically significant and medium and high dose group had statistically significance (P<0.05).[Conclusion] Fushen recipe may inhibit the occurrence of PMCs EMT by inhibiting TGF-β1 and regulating the expression of E-cadherin and α-SMA. |
