Establishment and evaluation of phlegm and blood-stasis syndromn on coronary heart disease model in mice

Abstract
    [Objective] In this study,a mouse model of coronary heart disease with phlegm and blood-stasis syndrome was established by coronary artery left anterior descending artery ligation combined with high-fat diet to simulate the pathological features of myocardial ischemia combined with atherosclerosis,so as to achieve the simulation of pathological changes in this syndrome.[Methods] Healthy SPF C57BL/6J mice were used in the control group and the sham operation group,and ApoE-/- mice were used in the blood stasis group,phlegm group and phlegm and blood-stasis group. The phlegm group and the phlegm and blood-stasis group were fed with high-fat diet for 12 weeks,and the other groups were fed with normal feed for 12 weeks. At the end of the 8th week,the left anterior descending coronary artery was ligated in the blood stasis group and the phlegm and blood-stasis group. The sham operation group was not ligated.[Results] The general state of each group of mice was observed. Echocardiography was used to detect serum lipid levels in mice. HE staining of heart tissue and oil red O staining of aortic tissue were performed to evaluate the phlegm and blood-stasis syndrome model of coronary heart disease.[Conclusion] ApoE-/- mice were treated with left anterior descending coronary artery ligation combined with high-fat diet for 12 weeks featured with the myocardial ischemia and atherosclerosis pathology,suggesting that this method can successfully establish the model of the coronary heart disease with phlegm and blood-stasis syndrome in mice.
